IBA LifeSciences StrGate Acptr Vctr pASG-IBA45
The pASG-IBA45 vector is designed for expression of recombinant proteins with an N-terminal Strep-tagII and a C-terminal His-tag in E. coli. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells, F1 and ColE1 origin for a high plasmid copy number, the inducible tetracycline promoter/operator for the regulated expression of proteins. Expression of the recombinant protein is induced by addition of anhydrotetracycline, which is a derivative of tetracycline that does not exhibit antibiotic activity. Please note that cloning into pASG-IBA Acceptor Vectors compulsorily requires the restriction enzyme Esp3I since no other MCS for the integration of a gene of interest is available. In addition to the direct cloning of the gene of interest into pASG-IBA vectors with Esp3I, another option via a so-called Entry Vector is possible.

Cre recombinase is used as a tool to genetically modify genes, such as to delete a segment of DNA flanked by LoxP sites in cells or experimental animals.By applying the mammalian codon usage to Cre recombinase, expression of Cre is improved in the mammalian cells. This improved Cre (iCre) gene also reduce the high CpG content of the prokaryotic coding sequence, thereby reducing the chances of epigenetic silencing in mammals.This AAV2 stock expresses an improved Cre (iCre) along with an eGFP under a control of the same CMV promoter. The Cre and eGFP was separated by a 2A peptides.

New England Biolabs, Inc. Lambda DNA (dam-) – 250 µg
Duplex DNA is isolated from bacteriophage lambda (cI857ind 1 Sam 7). The bacteriophage is grown in an E. coli host which is deficient in adenine methylase (dam-). Therefore, those deoxyadenosine positions which are methylated in a wild type E. coli host are not methylated in the dam- host. This DNA gives complete digests with restriction endonucleases that are sensitive to dam methylation. Lambda DNA (dam-) is 48,502 base pairs in length.
New England Biolabs, Inc. PhiX174 RF I DNA – 150 µg
This is the double-stranded, covalently closed, circular form of X174 RF I DNA (supercoiled). The molecular weight of X174 RF I is 3.50 x 10^6 daltons and it is 5,386 base pairs in length. The vast majority of the molecules are RF I, and the remainder are RF II, as determined by agarose gel electrophoresis.
Vector Biolabs v-raf-1 murine leukemia viral oncogene homolog 1 Adenovirus (Ad-RAF1)
The Raf kinases are important intermediates in signal transduction pathways. Raf-1, the prototype member of this gene family, is a 72-76 kDa cytoplasmic protein with intrinsic serine/threonine kinase activity, is broadly expressed in many cell types and is the cellular homolog of the viral oncogene, v-Raf. Other members of the Raf gene family include Raf-A and Raf-B. Raf-1 phosphorylates and thereby activates MEK (also designated MAP kinase kinase). Ras proteins bind Raf-1, but only when Ras is in its active GTP-binding state. This interaction results in Raf-mediated MEK activation. Two more recently described Raf-related proteins, Ksr-1 and Tak1 (TGFbeta-activated kinase), function as upstream regulators of Ras signaling pathways.
